In general terms the present invention proposes a method for calculating a biological age of a biological sample. The method comprises collecting the biological sample from a subject; preparing a single-cell RNA (scRNA) sequencing library from the biological sample, wherein preparing the scRNA sequencing library comprises a modified polymerization time and a modified purification process comprising two or more purification steps: identifying at least one type of genetic difference in the biological sample from the single cell RNA (scRNA) sequencing library; and gencrating a cell lineage tree that reflects a branching order of cells of the biological sample, wherein the branching order, with branch lengths included, of cells provides a data set for validating the biological age of the biological sample. The present invention also proposes a cell lineage tree based aging timer for calculating a biological age of a biological sample using the aforementioned method.
