本发明涉及药物领域，具体地公开了一种新型多荷载连接子的制备方法及其在偶联药物的应用。连接子以季戊四醇为母核，季戊四醇的三个羟基分别与两种不同的连接头结合，另外一个羟基与N‑长链的马来酰亚胺连接，构建出三荷载的新型连接子。本发明提出的连接子，三个连接头通过点击化学或环合反应等，实现与两种不同类型的效应分子连接；马来酰亚胺端与靶向配体连接，构建的偶联药物中载荷两种不类型的三个效应分子。
