本发明涉及环肽药物合成技术领域，具体涉及一种利用OaAEP1酶在硫酯辅助下合成镜像环肽的方法，首先运用以N芴甲氧羰基(Fmoc)固相多肽合成方法合成线性的镜像环肽硫酯前体，然后在OaAEP1酶的介导下硫酯前体即可生成镜像环肽。本发明首次在硫酯辅助下使用OaAEP1酶合成了镜像环蜂毒肽（D‑cMelittin），其具有高蛋白酶稳定性以及良好的抗菌活性。
