本发明提供一种特戈拉赞中间体的合成方法，所述特戈拉赞中间体合成方法包括：以1‑苄基‑4‑羟基‑2‑甲基‑1H‑苯并咪唑‑6‑羧酸为起始原料，依次经过酰胺化、与乙酰基进行酯化反应、催化氢化脱苄基，磺酸酯化，脱乙酰基保护得特戈拉赞中间体4‑羟基‑N,N,2‑三甲基‑1‑对甲苯磺酰基‑1H‑苯并[D]咪唑‑6‑甲酰胺。本申请采用先酰胺化再酯化反应，首先将高活性的羧基转化为稳定的酰胺，酰胺键在后续的酯化和氢化反应中都非常稳定，不会发生副反应。
