本发明公开了一种猪STING蛋白K61R突变体及其应用，所述突变体为猪STING蛋白的第61位赖氨酸突变为精氨酸，所述猪STING蛋白的氨基酸序列如SEQ ID NO.1所示。本发明首次鉴定猪STING蛋白的种属特异性关键位点K61，该位点为猪特有、区别于其他物种，通过调控蛋白稳定性及下游信号转导，对ASFV、PRV、HSV‑1等病毒复制发挥关键负向调控作用；同时，K61位点可直接作为猪抗病毒育种的核心分子标记与靶向位点，为提升抗病育种研究效率提供重要支撑。
